In vitro Experiment of Regeneration of Incised Meniscus after Application of Collagenase - a Preliminary Report -

Abstract
Purpose: To examine whether application of collagenase to the torn meniscus could promote the cure and proliferation of the chondrocyte.

Materials and Methods: After making the longitudinal tear to the lateral menisci obtained from 4 patients, 4% collagenase was applied to them for ten minutes in experimental group and not in control group, and then they were cultured in incubator for two weeks. They were stained with hematoxylin and eosin and histologically examined.

Results: In control group, incised gaps were still observed with sharp margin, and there were no difference between cells surrounding gaps and those of other area in shape and density, suggesting no evidence of regrowth. In experimental group, there were many small gaps in the extracellular matrix surrounding incision in all four cases. Although the density of cells from three menisci in experimental group seems to be similar to that of control group, partially increased number of cells were observed in some area compared to control group. There was marked increased density of cells and incised gap was not clear in one meniscus of experimental group. We thought that spindle cells found in dense area were the evidence of the proliferation of precursor cells of chondrocytes.

Conclusion: Collagenase applied to the torn meniscus for 10 minutes could weaken the extracellular matrix of meniscus partially , and application of collagenase to the torn menisci could proliferate the chondrocytes and be helpful for healing of torn menisci.
